I run vista64, and had the same problem at frist. All i did to fix the single player, is wait intill the client finished updateing. (meaning both the single player toggle and the multiplayer toggle were ready and active-ie= not grayed out). Then right click on the desktop launcher, and run as admin. It takes a min or two but launches fine. Been playing the singleplayer eversince with no problems as of yet. It did take 10-15min for the client to update, first time.
